Il mantenimento di un tessuto funzionale per tutta la vita richiede un continuo ricambio cellulare. Il tasso di ricambio è quindi attentamente bilanciato con le cellule malfunzionanti e morenti. Il ricambio cellulare fisiologico si basa principalmente sulle cellule differenziate terminali o staminali residenti. Le cellule terminali differenziate possono avere una tendenza alla replicazione che consente loro di propagare i precursori proliferanti. La maggior parte degli organi e dei tessuti ha le proprie cellule staminali o progenitrici che conservano la capacità di differenziazione. L’indirizzamento delle cellule staminali verso la destinazione dipende dalla loro nicchia e dal loro microambiente. Alcuni fattori esercitano il loro effetto sulle cellule staminali per guidare l’espressione di geni specifici che, a loro volta, promuovono la propagazione e il differenziamento delle cellule (Sanchez Alvarado e Yamanaka, 2014).
