O uso de células estaminais adultas para aplicações como engenharia de tecidos e terapia genética é uma das principais promessas da medicina regenerativa. No campo das células estaminais adultas, as células mesenquimais estromais da medula óssea (BMSCs) são candidatas muito promissoras. A capacidade de avançar na utilização destas células em terapia depende da criação de modelos mamíferos, necessários para avaliar a eficácia pré-clínica e padronizar os procedimentos. No entanto, as BMSC são um tipo de célula mais bem estudado e caracterizado em humanos do que em ratos e, embora recentemente tenham sido desenvolvidos numerosos protocolos de isolamento, ainda subsistem alguns problemas. Neste estudo, defino as condições para otimizar os rendimentos de culturas enriquecidas com progenitores específicos. Os resultados indicam que a escolha do meio utilizado para o isolamento e expansão das BMSCs murinas é extremamente relevante para enriquecer a cultura dos progenitores desejados. Este trabalho também sugere que níveis de expressão definidos de um marcador bem conhecido de células estaminais embrionárias indiferenciadas, nomeadamente Nanog, podem afetar a migração, invasividade e propriedades tumorigénicas das BMSCs e indica a necessidade de uma compreensão aprofundada da biologia destas células.
