Il costante miglioramento delle piattaforme di sequenziamento del DNA sta riducendo il tempo necessario per il processo di lettura e identificazione dei genomi a un costo sempre minore. Tuttavia, i sequenziatori presentano ancora alcune limitazioni, come ad esempio la dimensione dei frammenti di DNA che sono in grado di identificare. Pertanto, il DNA viene frammentato prima del sequenziamento e, dopo questa fase, riordinato in modo da poter rappresentare il genoma originale. Questo processo, noto come assemblaggio di genomi, richiede l’uso di diverse strategie in vitro e in silico. Negli ultimi anni sono state proposte diverse strategie per l’assemblaggio dei genomi, ma non esiste ancora un consenso sul miglior approccio da adottare. Recentemente, diversi genomi sono stati completati utilizzando una tecnica nota come mappatura ottica del genoma completo (WGM). Questa tecnica consente assemblaggi di alta qualità a partire da una mappa di restrizione ad alta precisione. La mappa di restrizione consente di orientare e ordinare i contig senza la necessità di un genoma di riferimento. La mappatura ottica consentirà l’avvento di una nuova era di assemblaggi genomici più rapidi, meno costosi e sempre più precisi.
