L’hypothyroïdie congénitale est une production insuffisante d’hormones thyroïdiennes chez le nouveau-né. Elle peut être due à un défaut anatomique de la glande, à une erreur innée du métabolisme thyroïdien ou à une carence en iode. On parle alors de crétinisme. L’hypothyroïdie congénitale (HTC) est une affection résultant de l’absence ou du sous-développement de la glande thyroïde (dysgénésie) ou d’une glande qui s’est développée mais qui ne peut pas produire d’hormones thyroïdiennes en raison d’un problème au niveau de la chaîne de production (dyshormonogenèse). Les bébés atteints de TCS ne peuvent pas produire suffisamment d’hormones thyroïdiennes pour répondre aux besoins de l’organisme. Le terme 'congénital' signifie que la maladie est présente à la naissance.
