La calreticulina (CRT) si occupa delle proteine mal ripiegate legandosi ad esse e contrassegnandole per la degradazione, in modo che solo le proteine correttamente ripiegate possano allontanarsi dal reticolo endoplasmatico. Si prevede che una proteina chinasi attivata da mitogeni a valle possa mediare questa fosforilazione. Pertanto, è stato utilizzato un trattamento con acido salicilico per potenziare l’espressione del gene CRT nel pomodoro. L’attività del sottogruppo ERK delle MAPK può essere inibita dagli inibitori ERK. L’inibitore ERK ha interrotto l’interazione fisica delle MAPK di tipo ERK e delle loro MAPKK a monte. La risposta delle proteine non ripiegate (UPR) causata dallo stress del RE è stata indotta dalla tunicamicina (TM) o dal ditiotreitolo (DTT). È stata condotta la clonazione dell’espressione di CRT in E.coli e l’espressione della proteina CRT è stata esaminata mediante SDS-PAGE e Western blotting. Sono stati esaminati anche due enzimi, la β-1, 3-glucanasi (PR2) e la glutammina sintetasi, sottoposti a trattamento di stress. Si spera che questo lavoro produca nuove conoscenze su diversi aspetti dei percorsi MAPK per facilitare una comprensione completa e molecolare del complesso sistema di difesa. Le scoperte getteranno le basi per strategie innovative basate sulla conoscenza per ridurre l’impatto delle malattie delle piante.
