Eine der wichtigsten Entwicklungen in der wissenschaftlichen Forschung der letzten Jahre war die Möglichkeit der In-vitro-3D-Zellkultur beim Menschen. Diese Technik ermöglicht wichtige Fortschritte in der Forschung, vor allem in der Krebsforschung, da 3D-Strukturen die Tumorumgebung besser imitieren können als 2D-Zellkulturen. In dieser Arbeit, die in zwei Kapitel unterteilt ist, präsentieren wir einen Überblick über Prostatakrebs, 3D-Zellkulturen und Methoden zur Auswahl von Liganden gegen Tumorzellen. Im zweiten Kapitel wird schließlich die Anwendung eines sehr effizienten Systems der 3D-Zellkultur erläutert, mit dem Ziel, RNA-Aptamere speziell gegen Prostatakrebszellen auszuwählen. Auf diese Weise haben wir den 3D Cell-SELEX entwickelt, mit dem Ziel, den Aptamer-Auswahlprozess zu innovieren und spezifischere Bindungsmoleküle auszuwählen, die effizient bei der Diagnose und Therapie von Prostatakrebs eingesetzt werden können.
