Die enzymatische Ligation von Peptid- und Proteinfragmenten stellt eine große Herausforderung dar. Unter anderem fehlen geeignete Biokatalysatoren, welche sowohl eine ausreichende Interaktion mit dem Peptidrückgrat aufweisen als auch die Knüpfung einer Peptidbindung katalysieren. Lars Franke untersucht die Möglichkeit, die hervorragenden Ligationseigenschaften der Surfactin-Thioesterase II auf anionisches Rattentrypsin II zu übertragen, welches seinerseits eine geeignete Interaktion mit dem Peptidrückgrat zeigt. Während die Katalysemechanismen beider Enzyme im Wesentlichen dem der Serinesterasen entsprechen, besteht ein Unterschied in der Architektur des Oxyanion-Lochs. Entsprechende Oxyanion-Loch-Mutanten des Trypsin wurden erzeugt, bezüglich ihrer katalytischen Eigenschaften untersucht und ihr Nutzen für enzymatische Ligationsreaktionen quantifiziert. Im Ergebnis konnte der Autor zwei Trypsinvarianten generieren, welche für die Peptidligation und die Proteinmodifizierung geeignet sind.Der AutorLars Franke ist in der Entwicklung von maßgeschneiderten Enzymen für die Peptidsynthese und Proteinmodifizierung am Charles-Tannford-Proteinzentrum in Halle tätig.
