L’enterite virale dell’anatra, causata dal virus dell’enterite dell’anatra (DEV), è una malattia acuta, contagiosa e letale. Il DEV è attualmente classificato nella sottofamiglia delle alphaherpesvirinae della famiglia degli herpesviridae. La DNA polimerasi, un prodotto della proteina UL30, riveste un’importanza fondamentale per il successo della replicazione virale nel gruppo dei virus alfa-herpesvirinae. L’enzima virale è coinvolto negli eventi iniziali dei processi di amplificazione genica. La proteina UL30 è altamente conservata tra gli herpesvirus a livello di sequenza aminoacidica. Si tratta di una delle proteine fondamentali per la replicazione del virus nella cellula ospite. Le sequenze di DNA che codificano per UL30 sono state identificate nel DNA genomico del DEV. In questo lavoro abbiamo sviluppato una proteina UL30 ricombinante del DEV a partire da un sistema procariotico. La proteina ricombinante espressa, ottenuta come proteina di fusione con tag istidina, è stata purificata mediante cromatografia di affinità. La proteina ricombinante purificata è stata confermata mediante SDS-PAGE e analisi western blot utilizzando sieri anti-DEV, che hanno rilevato una banda proteica da 80 kDa nel blot. La proteina ricombinante è stata successivamente dializzata e concentrata utilizzando filtri Amicon e la resa totale della proteina è risultata pari a 120 µg/ml.
