Acredita-se que os oncogenes retrovirais se formem através de erros na replicação do genoma do retrovírus após a sua integração em locais aleatórios no ADN do hospedeiro. O resultado final é um gene viral que é estruturalmente semelhante ao seu homólogo celular, mas persistentemente diferente na sua função. Por exemplo, o oncogene viral é quase idêntico ao gene de uma cadeia do fator de crescimento conhecido como fator derivado de plaquetas (pdgf). Os oncogenes celulares são altamente conservados na evolução.
