Lucena era hijo de Juan Ramírez de Lucena, embajador de los Reyes Católicos. Gracias a los viajes de su padre Lucena pudo viajar a Italia y Francia y conocer las innovaciones con respecto al ajedrez y al teatro. Nacido en Aragón, fue en sus años de estudiante de leyes en Salamanca cuando conoció a Juan del Encina. Entre ambos se desarrolló una amistad que iba a durar toda la vida. Lucena estaba trabajando en la imprenta de Nebrija en los años 1493-1495, donde le ayudó con ciertos términos para su diccionario de latín de 1495. Al parecer, Lucena intentaba traducir y editar en castellano libros en latín y así vemos aparecer: Mujeres ilustres en romance (1494) Fábulas de Esopo (1495), los libros de Aeneas Silvo Piccolomini (1496), Repetición de amores y Arte de Ajedrez (1497), Vita Beata (1499), El Jardin de nobles doncellas (1500). Poco después iba a dirigir y editar en Italia las nuevas cenas en la obra de Calisto y Melibea con la valiosa ayuda de su gran amigo Juan del Encina.
